Benjamin Franklin published an essay in the Pennsylvania Journal under the pseudonym American Guesser in which he suggested that the rattlesnake was a good symbol for the American spirit:

Metallica is obviously comprised of right wing extremist, if only they could be bought. Maybe even make a techno/rock mixed album.
This is A Song that is about the History of America and the Motto of the People of America.

And about the whole right wing thing, they have lots of other songs being hostile towards the "system" : Ride the lightning, Disposable heroes, and justice for all, eye of the beholder(especially about how corrupted freedom of speech have been made) to name some of them.
This song is simply about the revolutionary war and being proud of the americans who fought in it. Look at basically any of their others song about war and you'll see they aren't warmongers (One, For whom the bell tolls, disposable heroes, fight fire with fire)

Interpretation possibility #1 (assumed):
The song "Don't Tread On Me" could be a very patriotic song, because it is connected with America's "War of Independence".
Interpretation possibility #2 (assumed):
On the other hand it can be interpretated in the exact opposite way. In that aspect it could be a condemnation of blind patriotism. The Americans started out as the revolutionaries who just wanted to fight for their freedom and their country. Today they are supposed to be "The World's Police" who have to fight in almost every war all over the world. Probably the lyrics are close connected with the Gulf War I, because the song has been written around that time
