Depends what you mean by "supports". I highly doubt that there is any non-multi-billion-dollar software out there that is actually coded to utilize all available cores to the max efficiency - most seasoned programmers got no clue how to do it, since they never learned it 20 years ago. Having said that, on my quad-core I see all CPU at about the same level, but this workload distribution is done by the OS and not the software.
I have an i7 920 2.66GHz
Asus P6T Deluxe Motherboard
G.Skill 4GB DDR3 1333
Nvidia GeForce 9800 GT Superclocked Edition 512MB GDDR3
Corsair 750W PSU
The game runs flawlessly on highest possible settings and 1920x1080 resolution.

To simply answer the question by the OP, Yes it does, it uses all 4 of my cores when I run the game. Tested and confirmed with Riva Tuner+mods. And runs great other than I need to upgrade my 8800GT Video Card :P

I'd say it depends entirely on the size of your monitor.
You're buying pretty much the top of the line technology there is currently. I'm running at 1650x1080 on a 22" HPw2207 and a QX9550 @ 3ghz with a 8800GTX card and it runs just fine.
But yeah, it should last you a few years, although I would wait till Windows 7, and thus DX11 hardware arrives. I mean why buy a top of the line card now that may-may not support DX11 if you can wait 2~3 months to see what Nvidia has in store. (ATI announced a DX11 card already)
Thanks for the quick replys guys/Girls I guess ill be buying the PC i listed above... cant wait to try start playing!
Useing a Samsung SyncMaster 2253bw 21 monitor.
