I noticed in videos of character creation that you have to make certain choices about your character’s background and preferences. I assume the personal story will continue to present you with many more choices.
Do these choices have any effect on combat or build mechanics or are they simply for aesthetic and RPG purposes?

The personal story choices onlef affect your personal instance in your hometown. It would be unfair to have your atributes or combat mechanics affected by your choices on the personal story.
For example a permanent damage bonus you got by helping a priest of balthazar would be unfair vs someone who helped lyssa and all they got was a 5% discount in shops (all this example was made by me, this is not and wont be in game)
It's mostly for aesthetic / RPG purposes. The personal story is basically there for people who like more traditional quests. The decisions you make also effect your personal instance. So, once you do enough of the personal story, you could invite people into your personal instance, and show off how cool it is, or they could invite you into theirs and show you how different there's looks when compared to yours.
Other than that, people haven't spent a ton of time on this (as personal story coverage can very much spoil the experience), so we don't really have a good idea of just how different one will be to the next. It's kind of going to be a 'wait and see' type deal.
They can effect your Personal Story, but not your build choices. The Personal Story is an RPG experience with in GW2 that allows your character to follow a branching story line that is customized to the choices you make in character creation and then modified by some key choices you make in the story line itself.
This mode utilizes stylistic voice acted cut scenes, as well as instanced adventure content that is part of your story. You have to travel to the part of the world where each episode takes place and interact with a particular NPC or Object to enter that episode.
There is not enough story content to level all the way to cap, but it procides a nice adjunct to leveling via world PvE content and the story will tend to direct you to areas of the game appropriate for your progression. The item rewards are supposed to be nice and RPG fans will find this mode rewarding in it's own right, but it's completely optional. Players not interested can skip this content if they wish, but it would probably be a shame to do so.
